Epigenetic senescence and calcification of the aortic valve: a long journey from chromatin remodeling to nuclear mechanics

The calcification of the aortic valve is one of the most rapidly increasing cardiac pathologies. It is driven by an atherosclerosis-like inflammatory process and is mediated by transformation of the valve resident cells, the so-called valve interstitial cells into calcific nodule secretory cells. Several factors are responsible for pathologic evolution of the valve interstitial cells, including alterations in the epigenetic landscape. In the seminar I will present unpublished data linking cellular senescence and epigenetic alterations as a cause of valve interstitial cells calcification. I will also outline the possibility to interfere with calcification process by an epigenetic therapy. I will also describe the implications of epigenetic senescence on alterations of nuclear mechanics and chromatin accessibility.
