RING-Bait technology as a novel system to degrade pathological protein aggregates

Protein clustering and aggregation are fundamental cellular processes with distinct roles in health and disease. Clustering of proteins facilitates key functions like signal transduction, gene regulation, and enzymatic activity by organising molecular interactions. In contrast, protein aggregation results from misfolding or processing errors, often leading to cellular dysfunction and instability. These aggregates are often toxic, accumulating in cells and leading to various diseases, such as neurodegenerative disorders like Alzheimer’s and Parkinson’s. Removing these harmful protein aggregates is challenging but offers a promising approach to treating these diseases. In this seminar, I will introduce the “RING-Bait” technology, an innovative approach designed to specifically target and degrade pathological protein aggregates. This technology has successfully removed tau protein aggregates, a hallmark of Alzheimer’s disease, in cell models and mouse brains, leading to an improvement of motor function in transgenic mouse models. Furthermore, I will discuss how such targeted protein degradation strategies hold transformative potential for the treatment of protein-aggregation diseases, offering a wider application spectrum in various disorders.
