SMART SCISSORS & SMARTER ALGORITHMS: COMPUTATIONAL BIOLOGY AND CRISPR VS. CANCER

Abstract
I will present recent efforts from the Iorio Lab at Human Technopole to map genetic dependencies and therapeutic vulnerabilities in cancer cells through the integration of CRISPR-based perturbation screens and computational methods.

We develop and apply methods to predict and interpret how specific genomic alterations, including variants of unknown significance, create targetable weaknesses in tumor cells. By combining high-throughput CRISPR screening with multi-omics data and predictive algorithms, we identify context-specific dependencies, uncover mutation-induced “self-addictions” and discover opportunities for drug repositioning.

Together, these approaches demonstrate how “smart scissors” (CRISPR) and “smarter algorithms” can jointly advance the discovery of cancer vulnerabilities and support next-generation precision oncology.
