Prompted by large scale genomic initiatives, much research into cancer biology, and more specifically on drug resistance of cancer cells, have been focused in the past few years on the discovery of somatic genetic lesions in putative cancer genes, the definition of the underlying oncogenic mechanisms, and ultimately the identification of compounds specifically targeting rearranged genes. We posit that this approach has largely failed to improve the outcome of cancer patient. We argue that a more comprehensive elucidation of the evolutionary strategies deployed by cancer cells to increase their genetic and epigenetic landscape, at the single cell level, could finally uncover the wirings and mechanisms underlying cancer resistance.
