Pleiotropic control of B cell survival by p66Shc and its dysregulation in CLL

The Shc family of protein adaptors consists of four members, several of which expressed as different isoforms, that together control central cellular processes, including proliferation, survival, chemotaxis and apotosis. Here I will present on overview of the work of my group on p66Shc, a pro-apoptotic and pro-oxidant isoform of ShcA, that we have identified as a central player in the signaling pathways emanating from antigen and chemokine receptors in T and B cells. I will discuss the mechanisms by which p66Shc exerts a multilayered control on B cell survival and how these are dysregulated in chronic lymphocytic leukemia B cells, that are defective in p66Shc expression. I will also present unpublished evidence highlighting p66Shc as a new regulator of selective B cell autophagy.
