By training young scientists in setting up state of the art immunology tools and techniques (eg high throughput human monoclonal antibody production, single cell sorting, MHC tetramer technology, T cell epitope mapping, single cell intracellular cytokine assays, and somatic hypermutation analysis) in the lab at ICGEB, the goals of the ongoing research are to gain an in-depth understanding of human immunity dengue and chikungunya virus infections.
1. Using systems biology to define the molecular signatures of innate and adaptive immune responses during dengue and chikungunya infection.
2. To determine whether IgG Fc glycan composition is a determinant of antibody functionality and its impact on pathogenesis of disease.
3. To characterize the human B cell response during dengue and chikungunya infection – their functionality, specificity, isotype usage and affinity maturation.
4. To phenotypically and functionally characterize CD4 and CD8 T cell responses in dengue and chikungunya patients.
5. To generate human monoclonal antibodies that may provide novel avenues towards dengue and chikungunya therapeutics.
