NADPH Oxidases and Redox Regulation of Cardiac Remodelling

Oxidative stress is well recognized to be involved in cardiovascular pathophysiology.Beyond oxidative damage and the in activation of nitric oxide, redox signalling mediated by specialized ROS-generating enzymes – the NADPH oxidases (Noxs) – plays an important role in modulating cellular signalling pathways.The two Nox isoforms expressed in the heart,Nox2 and Nox4, have contrasting effects oncardiac remodelling.
Whereas Nox2 amplifies several signalling pathways that promote the development of heart failure, Nox4 has found to mediate protection against haemodynamic overload-induced remodelling and heart failure.This lecture will focus particularly on some of the mechanisms involved in these protective roles of Nox4 including modulation of the integrated stress response and novel effects on metabolism.
