High Throughput Screening

Equipment

The High-Throughput Screening Facility at the ICGEB is equipped with state-of-the-art instrumentation to perform genome-wide RNA interference screenings and is organized around three major workstations:

  • Liquid handling (Hamilton STARlet)
  • Multimode plate reading (PerkinElmer Envision)
  • High-content screening microscopy (Molecular Devices ImageXpress Micro)

Other small instruments available at the HTS facility include reagent dispensers (Thermo Multidrop), plate washer (Biotek ELx405) and plate sealer (ALPS).

 

 

Hamilton STARlet Liquid Handling Workstation

Technical specifications

  • 96-channel pipetting head 0.1-1000ul
  • 2 independent channels 0.1-1000ul
  • air displacement pipetting technology
  • 30 plate positions on-deck
  • gripping tool - CORE-gripper

This workstation is devoted to all tasks involving the handling of library stock plates including library ressuspension, dilutions, reformatting from 96- to 384-well plates, preparation of assay plates for screenings and cherry-picking.

Total handling capacity per unattended run: 60 microplates

 

PerkinElmer Envision Multimode Plate Reader

Technical specifications

  • dual technology: filters and monochromators
  • filters: excitation 405, 485, 544, 590nm; emission 535, 579, 635nm
  • monochromators: quadruple monochromators for absorbance and fluorescence (230-1000nm in 0.1nm increments)
  • dual-dispenser unit, temperature control
  • stackers for automated loading of microplates

This workstation is used for the high-sensitivity detection of absorbance, fluorescence (fluorescence intensity top and bottom, fluorescence polarization) and luminescence; the monochromators provide high flexibility in wavelength selection for absorbance and fluorescence measurements.

Molecular Devices ImageXpress Micro High-Content Screening Microscope

Technical specifications

  • 5 objectives (4x-40x)
  • 5 filter sets (DAPI, FITC, TexasRed, Cy3, Cy5)
  • integrated with ThermoFisher Catalyst Express robotic arm for automated loading of microplates

This workstation is dedicated to the automated acquisition of fluorescence images from 96- and 384-well plates, thus allowing the quantitative analysis of cellular and subcellular events (e.g. cell cycle, cell health, gene expression, nucleo-cytoplasmic translocation, receptor internalization, spot detection, tube formation).
